Introdução: O sucesso nos procedimentos adesivos em odontologia manifesta-se por meio de restaurações duradouras. Contudo, a instabilidade da interface adesiva ocorre principalmente devido à degradação hidrolítica dos monómeros adesivos na camada híbrida ao longo do tempo, e à limitada capacidade dos adesivos em penetrar completamente na matriz de colagénio. Tais fibrilas de colagénio desmineralizadas e desprotegidas, subjacentes à camada híbrida, encontram-se expostas à proteólise provocada pelas enzimas hospedeiras. As metaloproteinases de matriz (MMPs) são enzimas capazes de degradar o colagénio na camada híbrida, contribuindo assim para a redução da longevidade das restaurações adesivas. Objetivo: Avaliar a eficácia de alguns inibidores da MMP na força de adesão dentinária. Metodologia de pesquisa: Foi realizada uma revisão de 9 artigos, no período de 2013 a 2023 (últimos 10 anos) foram utilizadas as bases de dados Medline, Scopus, B-on, sendo as palavras-chave: metaloproteinases, matriz de colagénio, adesão e dentina. Resultados: Os resultados foram agrupados com base nos meios utilizados para a inibição da MMPS: Baicaleína, Clorexidina, Reticulantes de Colagénio, Kaempferol, Proantocianidina, Inibidores Naturais vs Inibidores Sintéticos, Zinco, Terapia Fotodinâmica, Inibidor MMP8-I. Conclusões: Todos os agentes inibidores de MMPs avaliados nesta investigação apresentaram melhorias na microtração, sobretudo a longo prazo. Contudo, é imperativo realizar mais estudos com o intuito de estabelecer um protocolo clínico aceitável.

However, the instability of the adhesive interface occurs mainly due to the hydrolytic degradation of adhesive monomers in the hybrid layer over time and the limited ability of adhesives to fully penetrate the collagen matrix. These demineralized and unprotected collagen fibrils underlying the hybrid layer are exposed to proteolysis by host enzymes. Matrix metalloproteinases (MMPs) are enzymes capable of degrading collagen in the hybrid layer, thereby reducing the longevity of adhesive restorations. Objective: To evaluate MMP inhibitors on dentin adhesion. Research methodology: A review of 9 articles was carried out from 2013 to 2023 (last 10 years) Medline, Scopus, B on databases were used, the keywords being: metalloproteinases, collagen matrix, adhesion and dentin. Results: The results were grouped based on the means used for MMPS inhibition: baicalein, chlorhexidine, collagen cross linking agents, kaempferol, proanthocyanidin, natural vs synthetic inhibitors, zinc, photodynamic therapy, MMP8 I inhibitor. Conclusions: All MMP inhibitors assessed in this investigation demonstrated improvements in microtensile strength, particularly over the long term. Nevertheless, it is imperative to conduct further studies with the aim of establishing an acceptable clinical protocol.
