A necessidade de novos fármacos para as atuais patologias impulsionou a indústria farmacêutica na procura de novas aplicações para os fármacos já existentes
Um potente inibidor da fosfodiesterase-5, o chamado de Sildenafil, foi descoberto há cerca de 15 anos e cuja principal indicação é no tratamento da disfunção erétil.
Apesar da sua principal indicação o conhecimento dos mecanismos associados aos inibidores de fosfodiesterase-5 tem levado ao estudo de novas aplicações para esta substância. Sabendo que o seu efeito no metabolismo humano é conhecidamente vasodilatador, o uso em outras patologias além da disfunção erétil tem sido testado com sucesso.
Atualmente, o Sildenafil já tem sido utilizado no tratamento de outras patologias, nomeadamente, na hipertensão pulmonar devido à sua ação como vasorelaxante para a artéria pulmonar. No entanto os estudos revelam que este poderá ser utilizado em outras patologias desde as cardiovasculares até às doenças neurodegenerativas.
Este trabalho de revisão tem como objetivo principal realizar uma revisão bibliográfica acerca das aplicações, off label do Sildenafil e como estas poderão num futuro próximo contribuir no tratamento de diversas patologias.
The need for new drugs to current conditions boosted the pharmaceutical industry to seek new applications for existing drugs A potent inhibitor of phosphodiesterase-5, called Sildenafil, was discovered about 15 years and whose main indication is for the treatment of erectile dysfunction. Despite its main indication the knowledge of the mechanisms associated with phosphodiesterase-5 inhibitors has led to the study of new applications for this substance. Knowing that their effect on human metabolism is known to be a vasodilator, use in other diseases of erectile dysfunction has been successfully tested. Currently, Sildenafil has been used in the treatment of other pathologies, in particular pulmonary hypertension due to its action as a vasorelaxant to the pulmonary artery. However studies show that it may be used in other diseases from cardiovascular to neurodegenerative diseases. This review study aims to review literature about the applications, off label of Sildenafil and how they can contribute in the near future in the treatment of various pathologies.
