Introduction: Botulinum toxin, has a relatively well known mechanism of action. It is used for several procedures that have proven useful in various fields of medicine. Objectives: To understand how the evaluation of variations in muscle contraction patterns are crucial to therapeutic success in Oral and Facial Harmonization. The importance of anatomical knowledge of the facial muscles and the physiology of muscle contraction is naturally highlighted. Methodology: A literature review was conducted based on articles published online in the last ten years. Articles were searched in the PubMed, Bon, Elsevier, and Science Direct databases. The criteria used to include the publications were (a) presence of the descriptors used in the search in the title or abstract; (b) articles published in Portuguese, French, Spanish or English; (c) open access articles; (d) dissertations and theses. The exclusion criteria were (a) duplicate articles; (b) articles whose full content did not meet the proposed theme; (c) articles with texts not made completely available. Conclusion: Although botulinum toxin has a diversified and safe field of action in several medical areas, the knowledge of muscle contraction patterns is fundamental to optimize clinical protocols and determine therapeutic success.
